This conference poster presents the generation of hepatocyte-like cells (YBLiHepato) derived from a healthy human induced pluripotent stem cell (hiPSC) line to address the limited availability of primary human hepatocytes for liver research. The cells were sequentially differentiated into definitive endoderm, hepatic progenitor cells, and functional hepatocyte-like cells, demonstrating successful lineage-specific development.

Comprehensive characterization using FACS, qRT-PCR, LC-MS/MS, and functional assays confirmed the expression of key hepatic markers, active cytochrome P450 enzyme activity, lipid metabolism capability, and urea production. These findings highlight the potential of YBLiHepato cells as a reliable platform for drug metabolism studies, hepatotoxicity testing, and translational liver research.
